چکیده

Since interferon' per se shows little promise as a prophylactic or therapeutic agent, interest has shifted to a search for acceptable inducers by which the body might be stimulated to make its own interferon. Shope2 demonstrated that a substance which was derived from Penicilliumfuniculosum and which was called helenine induced resistance to Semliki Forest and to Columbia SK virus infections in mice. Lewis et al.3 fractionated helenine and prepared a factor active in the mouse assay which exhibited properties of a nucleoprotein. Rytel, Shope, and Kilbourne4 demonstrated that helenine elicited a viral inhibitor in cell cultures and in mice which exhibited properties similar to those of interferon. In commenting, they stated that "the active principle in helenine is still a matter of conjecture since it has not been obtained in a sufficiently pure form to permit definite identification." Studies in our laboratories during the past several years have been directed toward the discovery of an interferon inducer which would be worthy of clinical evaluation. It was found that certain ribonucleic acids were highly active in inducing interferon and host resistance but were dependent upon (a) freedom from inhibitory protein and (b) multistrandedness of the RNA. Ribonucleoproteins and single-stranded nucleic acids were inactive. This series of papers describes the purification or synthesis and characterization of three kinds of multistranded RNA active in inducing interferon and host resistance. The present report describes the isolation and characterization of a double-stranded RNA from extracts of Penicillium funiculosum (helenine) which, when freed of protein, is a highly active inducer of interferon and host resistance. The substance is referred to as HeI-RNA. Subsequent papers will present data showing high level interferon and host resistance-inducing activities of multistranded complexes of synthetic polynucleotides5 and of double-stranded RNA of reovirus origin.6

Interferon, double-stranded RNA, and protein phosphorylation.

We reported earlier that the addition of double-stranded RNA and ATP increases the endonuclease activity more in an extract of Ehrlich ascites tumor cells which have been treated with an interferon preparation than in a comparable extract from control cells. We report here that the addition of double-stranded RNA to an extract from Ehrlich ascites tumor cells which have been treated with an int...

Interferon Resistance of Hepatitis C Virus Genotypes 1a/1b: Relationship to Structural E2 Gene Quasispecies Mutations

Hepatitis C virus (HCV) envelope glycoprotein-2 (E2) inhibits the interferon (IFN)–induced, double –stranded RNA activated protein kinase (PKR) via PKR eukaryotic initiation factor-2α phosphorylation homology domain (PePHD). Present study examined the genetic variability of the PePHD in patients receiving interferon therapy. The PePHD region from HCV genotype 1a/1b infected patients receiving I...

Interferon-induced inhibition of protein synthesis in L-cell extracts: an ATP-dependent step in the activation of an inhibitor by double-stranded RNA.

The translation of encephalomyocarditis virion RNA in extracts from interferon-treated L-cells is inhibited by the addition of double-stranded RNA (dsRNA) at 400 ng/ml. A similar inhibition in response to dsRNA is seen in control cell extracts supplemented with small amounts of a postribosomal supernatant fraction from interferon-treated cells (interferon cell sap): Neither interferon cell sap ...

Nonviral Interferon Inducers

Interferon production can be stimulated by a great variety of microbial and nonmicrobial agents other than viruses. The nonmicrobial inducers can be divided into polyanions, mitogens, and a miscellaneous category including the various endotoxins and antibiotics. The polyanions appear to require a stable, high molecular weight backbone and a high density of free anionic groups whether they are p...

The dengue virus conceals double-stranded RNA in the intracellular membrane to escape from an interferon response

The dengue virus (DENV) circulates between humans and mosquitoes and requires no other mammals or birds for its maintenance in nature. The virus is well-adapted to humans, as reflected by high-level viraemia in patients. To investigate its high adaptability, the DENV induction of host type-I interferon (IFN) was assessed in vitro in human-derived HeLa cells and compared with that induced by the...
